细胞色素p450对大鼠心肌缺血 再灌注损伤的影响 |

摘 要 目的:通过观察细胞色素p450抑制剂对大鼠在体心肌缺血/再灌注损伤的影响,了解细胞色素p450与心肌缺血/再灌注损伤的关系,并初步分析作用机制。方法:观察给予细胞色素p450抑制剂或11, 12-EET后,大鼠在体心肌缺血/再灌注室性心律失常、心肌梗塞范围及超微结构的变化,及TEA与glibenclamide对EETs作用的影响。结果:细胞色素p450抑制剂clotrimazole(0.9 mmol/L)和SKF525A(0.54 mmol/L)减轻、而外源性11, 12-EET加重缺血/再灌注心律失常、心肌梗塞范围及心肌超微结构病变;TEA与glibenclamide可拮抗11,12-EET的作用。结论:细胞色素p450参与了心肌缺血/再灌注损伤;其作用与EETs释放并开通钙敏感性及ATP敏感性钾通道有关。 主题词 细胞色素P450;心肌;局部缺血;再灌注损伤
Effects of cytochrome p450 on myocardial ischemia/reperfusion injury in rats

Abstracts AIM: To study the effects of cytochrome p450 on myocardial ischemia/reperfusion injury and the mechanisms. METHODS: Observing the changes of ischemia/reperfusion arrhythmias , myocardial ultrastructure injury and myocardial infarct size after cytochrome p450 inhibitors or 11, 12,-EET(epoxyeicosatrienoic acids) and the influence of TEA (tetrae thylammonium) and glibenclamide on them. RESULTS: The cytochrome p450 inhibitors clotrimazole (0.9 mmol/L) and SKF525a (0.54 mmol/L)decreased but 11,12-EET increased the ischemia/reperfusion arrhythmia, myocardium infarct size and ultrastructure injury; the effect of 11,12-EET could be reduced by TEA and glibenclamide. CONCLUSION:Cytochrome p450 might relate to myocardial ischemia/reperfusion injury and the action of it may be through releasing EET and opening the KATP and KCa . 近年来,细胞色素p450单氧化酶及其花生四烯酸代谢产物环-二十碳三烯酸(epoxyeicosatrienoic acids, EETs)对心血管的影响引起了人们的兴趣。有报道在心肌缺血/再灌注时,心肌中细胞色素p450含量及活性均有所提高[1,2],这对于心肌缺血/再灌注损伤的意义如何尚不清楚;还有人指出,细胞色素p450代谢产物EETs可加重离体心脏再灌注后心功能障碍[3],但EETs对在体心肌缺血/再灌注损伤有何影响,尤其是对心律失常和心肌结构有何影响,尚未见报道。 本文旨在观察抑制细胞色素p450对大鼠在体心肌缺血/再灌注损伤的影响,并对其作用机制进行初步分析。
材料与方法
