大鼠主动脉内膜球囊剥脱后再生内皮细胞表型改变的研究 |

摘 要 目的:探索大鼠主动脉内膜球囊剥脱后,内皮修复过程中再生内皮细胞结构和功能的改变。方法:大鼠主动脉内膜球囊剥脱2周和6周对内膜进行形态学观察,同时测定血管反应性、亚硝酸盐含量和NOS的活性。结果:剥脱2周和6周后,再生内皮细胞不仅结构与正常内皮细胞不同,而且血管再生内皮依赖性舒张反应减弱、NO的释放明显减少、血管组织iNOS和cNOS活性均比正常降低;剥脱6周后的iNOS比剥脱2周时有所增加,但cNOS则无明显差别。结论:实验提示在大鼠主动脉内膜剥脱后的内皮细胞再生增殖过程中,再生内皮细胞功能发生了表型改变,从而不能有效地抑制平滑肌细胞的增殖和迁移;提示再生内皮细胞功能的功能不足或紊乱可能是再狭窄病变形的重要因素之一。 主题词 再生;内皮;一氧化氮;大鼠

Abstract AIM: To investigate the changes of structure and function in regenerated endothelial cells (REC) after the denudation of endothelium in rat aortic artery.METHODS:After the denudation of endothelium in rat artery by balloon, REC structure was revealed by H E staining, the reactivity of endothelial dependent relaxation (EDR) in vessels, release of endothelial nitric oxide(NO) and vascular nitric oxide synthase (NOS) were investigated.RESULTS: REC structures were different from normal endothelial cell; EDR in vessels after denudation for 2 weeks and 6weeks decreased, and release of endothelial nitric oxide(NO) decreased companied with inhibited vascular inducible nitric oxide synthase(iNOS) and constructional nitric oxide synthase(cNOS).CONCLUSION:Phenotypic shift of REC during neointima formation after endothelial removal by balloon-injury, and dysfunction of REC might enhance neointimal impairment during retenosis formation after angioplasty.
