胎肝提取物对培养中4株肝癌细胞增殖和分化的影响 |

摘 要 目的:观察4月胎肝提取物对QGY-7703、BEL-7402、SMMC-7721人肝癌细胞和Hep A小鼠肝瘤细胞增殖和分化的影响。方法:从4月胎肝中制备提取物,采用[3H]-TdR掺入法和MTT比色法。 结果:100~500 mg/L的胎肝提取物能明显抑制QGY-7703、BEL-7402、SMMC-7721、Hep A等肝癌细胞的DNA合成和脱氢酶活力。对DNA合成的半数抑制浓度(IC50)分别为346.93、 151.18、 182.37、231.03mg/L;对脱氢酶活力的抑制弱于对DNA合成的抑制,IC50分别为1318.1、1599.2、793.8、813.1mg/L。结论:胎肝提取物能够抑制上述4种细胞增殖和促进分化。 主题词 肝细胞瘤;生长抑制物;氧化还原酶类;细胞分化
Effect of embryo hepatic extracts on proliferation and differentiation of 4 lines of hepatoma cells in culture

Abstract AIM: The effects of embryo hepatic extracts (EHE) on the proliferation and the differentiation of QGY-7703, BEL-7402, SMMC-7721 human hepatoma cell lines and HepA murine hepatoma cell were investigated. METHODS: EHE was extracted from 4-month human embryo,[ 3 H]-TdR incorporation and MTT colorimetric assays were performed. RESULTS: 100~500 mg/L of the extracts can apparently inhibit the DNA syntheses and the dehydrogenase activities of these cells. The half inhibitory concentration(IC50) of DNA syntheses of QGY-7703, BEL-7402, SMMC-7721, HepA hepatoma cells were 346.93, 151.18, 182.37 and 231.03 mg/L. The inhibitory effect of EHE on the dehydrogenase is smaller than that of these cells on DNA syntheses; the IC50 were 1318.1, 1599.2, 793.8, 813.1mg/L respectively. 已有研究表明,从4月胎肝中制备的提取物可以抑制BEL-7402细胞的增殖,形态上能促进BEL-7402细胞的分化[1]。胎肝提取物能否抑制其它肝癌细胞的生长和促进分化一直是我们关注的问题,为此我们选4株不同来源的肝癌细胞,观察胎肝提取物对4株肝癌细胞的影响。
材料和方法
