LMP2与LMP7基因多态性与强直性脊柱炎并发虹睫炎易感性关系的研究 |

【摘 要】 目的 研究LMP基因多态性与强直性脊柱炎并发虹睫炎发病的关系。方法 应用PCR对正常人和病人进行HLA-B27检测以及LMP2和LMP7扩增。CfoI进行限制性酶切图谱分析。结果 强直性脊柱炎有虹睫炎(AS＋AAU)病史以及单纯虹睫炎(AAU)患者LMP2基因BB纯合型较正常人以及强直性脊柱炎(AS)患者明显增高(P＜0.05)，AS＋AAU患者BB型OR＝3.6，AAU患者BB型OR＝5.83。LMP7基因多态性无明显差别。结论 在我国汉人中，LMP2基因多态性与AS＋AAU以及AAU发病存在明显相关关系。

Association of the polymorphism of LMP2 and LMP7 genes with the susceptibility to acute anterior uveitis in patients with ankylosing spondylitis

【Abstract】 Objective To evaluate the polymorphism of LMP2 and LMP7 genes in patients with ankylosing spondylitis (AS) and /or acute anterior uveitis (AAU).Methods HLA-B27 typing was performed and LMP2 and LMP7 amplified fragment length polymorphisms were determined after PCR and digestion with CfoI restriction enzyme.Results There was a significantly increased LMP2BB genotypic frequency in patients with AS＋AAU (85.7%) and AAU (87%) when compared with controls (53.3%) and AS patients (62.5%) (P＜0.05)，but no difference in LMP7 genotype.Odds ratio (OR) relating LMP2 BB and AAU was 5.83 and OR for AS＋AAU was 3.6.Conclusion The polymorphism of LMP2 gene is strongly associated with the development of AAU in AS patients and uncomplicated AAU patients.

强直性脊柱炎(AS)与HLA-B27基因存在强相关关系。在国人中AS与HLA-B2704相关性最强〔1〕。而HLA-B27阳性人群中，AS发病率及发病类型的家族倾向性又说明除了HLA-B27以外一定还存在其他的基因或因素在起作用，目前已经发现与HLA-Bw60以及HLAⅡ类分子基因等有关，但不同人种作用可能不同〔2〕。本研究旨在探讨LMP基因多态性与我国汉人AS发病的关系。
1 资料与方法
1.1 病人
本组患者共108例，均为汉族。AS符合修订的纽约标准，AS发生虹膜睫状体炎(以下简称虹睫炎)的患者(AS＋AAU)既往由眼科医师确诊。单纯虹睫炎(AAU)病人由我院眼科医师检查确诊。正常对照组血标本取自志愿者，见表1。
