骨关节炎关节软骨及游离体中胶原 型的表达 |

摘 要:目的 进一步了解与骨关节炎相关的关节软骨及游离体软骨细胞表型改变。方法 通过组化和免疫组化方法,观察纤维性胶原Ⅰ、Ⅱ、Ⅲ型在人正常和骨关节炎关节软骨及游离体中的表达。结果 在10例骨关节炎关节软骨标本中,有9例可在关节软骨的中层和深层的软骨细胞中显示强烈的Ⅱ、Ⅲ型胶原蛋白的阳性表达信号,而Ⅰ型胶原表达蛋白只在中下层及深层观察到,表达强度较Ⅲ型弱,表达区域部分与Ⅲ型重叠。在10例骨关节炎游离体标本中,均可在游离体内部软骨细胞及基质中显示强烈的Ⅲ型胶原蛋白阳性表达信号。相同标本中Ⅰ型胶原蛋白仅在游离体内部软骨细胞中有表达,表达的阳性信号相对较低,未见明显的Ⅱ型胶原表达。在正常关节软骨中,Ⅱ型胶原的表达的信号较弱,未见Ⅰ、Ⅲ型胶原蛋白表达信号。结论 在骨关节炎关节软骨及游离体中,一定区域内的软骨细胞表型发生了改变,表达合成了Ⅲ型胶原和Ⅰ型胶原,可能在骨关节炎发病机制中发挥重要作用。 关键词:软骨,关节; 胶原; 免疫组织化学; 关节游离体; 骨关节炎
Expression of collagens Ⅰ,Ⅱ and Ⅲ in chondrocytes of human osteoarthritic cartilage and loose body

Abstract:Objective To evaluate phenotypic changes of chondrocytes related to the disease.Methods Normal and osteoarthritic human articular cartilage and loose body were investigated by histochemistry and immunohistochemistry for expression patterns of the fibrillar collagens type Ⅰ,Ⅱ and Ⅲ.Results In 9 out of 10 samples,a defined subset of chondrocytes in the middle and deep zone of osteoarthritic cartilage showed significant levels of cytoplasmic protein of collagens Ⅲ and Ⅱ,whereas moderate signals of collagen Ⅰ protein were found in the lower middle and deep zone,partially overlapping with the zone of collagen Ⅲ protein-expressing cells.Collagen type Ⅲ expression was detected in 10 cases of loose bodies and was primarily located in the chondrocytes and matrices of loose bodies.As to collagen type Ⅰ expression,it was weakly positively stained in the chondrocytes of loose bodies.In normal articular cartilage,no detectable levels of cytoplasmic protein for collagens Ⅰ and Ⅲ,and weak expression level of collagen Ⅱ,were seen.Conclusion These results indicate a phenotypic alteration in a defined subset of chondrocytes on conditions of diseased cartilage and loose bodies,expressing and synthesizing collagen type Ⅲindependent of collagen typeⅠ,[1] [2] 下一页 上一个医学论文: 狼疮肾炎患者血清sFas和sFasL的变化 下一个医学论文: 男性与女性白塞病的对比研究
