雷公藤多甙体外对成骨细胞功能表达的影响 |

摘 要:目的 观察雷公藤多甙(MT)体外对成骨细胞(OB)功能表达的影响,以进一步探讨雷公藤(TW)致女性骨质疏松的机制。方法 分离新生SD大鼠头盖骨OB进行原代培养,经5~6 d至汇合,传代后加入不同浓度MT,用对硝基苯磷酸法和考马斯亮蓝法作碱性磷酸酶(ALP)比活性测定,茜素红染色法作钙化结节计数测定OB矿化能力。结果 培养6 d,MT 1 μg/ml起显著抑制OB ALP比活性(P<0.05);培养2周,MT 10-4 μg/ml起显著抑制OB矿化能力(P<0.05);药物对OB功能表达的抑制程度与其剂量密切相关(P<0.05,P<0.01)。结论 MT显著抑制OB功能表达且呈剂量依赖性。TW对OB的直接抑制作用,是其导致药物性骨质疏松的一个重要原因。 关键词:雷公藤; 雷公藤多甙; 成骨细胞
Effects of multiglucoside tripterygii on the function of osteoblasts in vitro

Abstract:Objective The effects of multiglucoside tripterygii (MT) on the function of osteoblasts were studied in order to further explore the mechanism of osteoporosis induced by Tripterygium wilfordii (TW).Method Osteoblasts were obtained from the calvariae of new-born SD rats and were cultured with MT of various concentrations.The effects of MT on alkaline phosphatase (ALP) activity and mineralization ability of osteoblasts were detected.Results 1 μg/ml MT significantly suppressed ALP activity,and 10-4 μg/ml MT inhibited mineralization ability of osteoblasts remarkably.These effects were significantly correlated with MT concentrations.Conclusion It is indicated that MT has direct suppressive effects on the function of osteoblasts, which may play an important role in the pathogenesis of osteoporosis induced by TW. 临床研究在整体水平上证实了长期应用雷公藤(TW)有致女性SLE患者骨质疏松的副作用,且随药物剂量的增加骨质疏松程度加重[1]。体外实验进一步证实雷公藤多甙(MT)可直接抑制成骨细胞(OB)增殖且呈剂量依赖性[2]。本研究进一步用对硝基苯磷酸法和考马斯亮蓝法作碱性磷酸酶(ALP)比活性测定,茜素红染色法作钙化结节计数测定OB矿化能力,以检测MT体外对OB功能表达的影响,在细胞水平上继续探讨TW致女性骨质疏松的机制。
1 材料与方法
1.1 动物 新生雌性Sprague-Dauley (SD)大鼠,放射医学研究所动物房提供。沪医实验动物合格证:34。 1.2 药物 雷公藤多甙(MT)由中国医学科学院皮肤病研究所郑家润教授提供,以DMSO助溶,PSB液按10倍稀释。 1.3 试剂
