大鼠血管球囊成形术后血管局部纤连蛋白的改变 |

【摘要】 目的 探讨细胞外基质在血管成形术后再狭窄中的作用。方法 在大鼠胸主动脉球囊损伤模型上,应用免疫组化和原位杂交的方法,观察纤连蛋白(Fn)在损伤后不同时间点和位置的蛋白和mRNA的表达。结果 Fn在球囊损伤后第3天开始表达,第7天进一步增加,14天达高峰,30天仍持续高表达,在新生内膜及近内膜的中层最为明显。结论 Fn在球囊损伤后异常增加,为新生内膜形成的重要原因,可能在成形术后再狭窄形成中起重要作用。 【关键词】 细胞外基质 纤连蛋白类 血管成形术,气囊

Methods The time and spatial course of mRNA and protein expression of fibronectin were exa-mined on the model of balloon angioplasty of the thoracic aorta in the rat with the combination of in situ hybridization and immunohistochemistry methods. Results The expression of fibronectin began at the third day after balloon injury and continued to increase at the seventh day, and reached the maximum from two weeks to one month after injury, which was most significance on the medial near basement membrance and neointima. Conclusion The variation of fibronectin may play a very improtant role in the migration of vascular smouth muscle cells, formation of neointima and restenosis after balloon angioplasty. 【Key words】 extracellular matrix fibronectins angioplasty, balloon
近年来,研究发现经皮腔内冠状动脉成形术(PTCA)后细胞外基质(extracellular matrix, ECM)大量合成和积累,在再狭窄过程中起重要作用[1]。纤连蛋白(fibronectin,Fn)是一种重要的细胞外基质成份、在组织的分化、胚胎的发育及组织损伤后的修复过程中起重要作用[2]。为此,我们在大鼠胸主动脉球囊损伤模型上,观察Fn的动态变化过程,以探索其在再狭窄形成中的作用。
材料与方法
1.动物:40只雄性Wistar大鼠,重350 g左右,购自北京医科大学动物室。 2.动物模型的制备:40只大鼠随机分成5组(n=8),正常对照8只,32只用5%戊巴比妥钠0.3 ml腹腔注射麻醉。由颈部分离左颈总动脉,用改良的2F Forgarty球囊导管从左颈总动脉插入胸主动脉至膈肌,以0.5~1.0 ml生理盐水充盈球囊至直径0.5~0.7 cm,反复回拉导管9次,拔出球囊,缝合伤口,常规饲养。分别于术后3、[1] [2] [3] 下一页 上一个医学论文: 局部释放肝素的生物降解性支架的初步实验研究 下一个医学论文: 电子束CT测定的冠状动脉钙化与冠心病危险因素的关系
