系统性红斑狼疮凋亡相关基因Fas sFasmRNA表达研究 |

摘 要:目的 从转录水平研究系统性红斑狼疮患者凋亡相关基因Fas、sFas表达情况,探索凋亡在SLE发病中的作用。方法 利用荧光标记RT-PCR半定量方法,以β-actin为内参照,对50例SLE、24例RA和24名正常人外周血白细胞凋亡相关基因Fas、sFas mRNA表达水平进行了检测。结果 SLE Fas、sFas表达均高于正常对照(3.2±1.8 vs 2.1±0.6,P<0.001和1.2±0.7 vs 0.58±0.27,P<0.000 1),并且表达量与病情活动度相关(相关系数分别为0.521 8,P=0.007和0.418 8,P=0.037),而RA仅sFas表达增高(0.9±0.5 vs 0.58±0.27,P<0.01)。结论 SLE患者外周血白细胞凋亡相关基因表达存在异常,提示凋亡在SLE发病中可能具有一定作用和意义。 关键词:红斑狼疮,系统性; 基因表达; 聚合酶链反应; Fas基因; 细胞凋亡
Expression of Fas and sFas genes in systemic lupus erythematosus patients

Abstract:Objective To investigate the mRNA expression of Fas and sFas genes in SLE patients and their roles in the pathogenesis of SLE.Method Fifty SLE patients,24 RA patients and 24 normal controls were chosen and the expression of Fas and sFas genes of peripheral white cells were detected by fluorescence-based RT-PCR semi-quantitative method.Results ① mRNA expression of Fas and sFas in SLE patients were significantly higher than that in normal controls (3.2±1.8 vs 2.1±0.6,P<0.001 and 1.2±0.7 vs 0.58±0.27,P<0.000 1,respectively),and relevant to activity of SLE,r=0.521 8,P=0.007 and r=0.418 8,P=0.037).② In RA patients,sFas was higher than that in normal controls (0.9±0.5 vs 0.58±0.27,P<0.01),but expression of Fas had no statistical difference.Conclusion The expression of Fas and sFas genes are abnormal in SLE and apoptosis may play certain role in the pathogenesis of SLE.
