【摘要】 目的 观察人重组胰岛素样生长因子-I(rhIGF-I)对STZ诱发的糖尿病大鼠心脏机械功能的作用,并和牛初乳短链胰岛素样生长因子-I(BC-tIGF-I)相比较。方法 STZ 55mg/kg空腹腹腔注射(IP)制备糖尿病大鼠模型,随机分为四组:DM-C组,生理盐水0.1ml/d IP;DM-L组,BC-tIGF-I 250ng.kg-1.d-1 IP;DM-H组,BC-tIGF-I 750ng.kg-1.d-1 IP;DM-rh组,rhIGF-I 750ng.kg-1.d-1 IP;并设正常对照组,每组5只。实验持续6周。心功能用室内导管法测定,观察指标有:室内压最大值(Peak)、舒张末期室内压(EDP)、室内压上升段最大变化率(dp/dt)、室内压下降段最大变化率(-dp/dt)。同时测定尾尖血血糖(BG)、血清果糖胺(FMN)、甘油三酯(TG)、胆固醇(Ch)。结果 与正常大鼠相比,DM-C组血清FMN、TG明显升高,同时Peak降低41.4%,dp/dt、-dp/dt绝对值分别降低40.3%、58.5%,EDP升高〔(2.00±0.62)mmHg vs (-0.44±0.26)mmHg, P<0.01〕。DM-L、DM-H、DM-rh组尾尖血BG、血清FMN、TG明显低于DM-C,但仍高于正常。各组心功能指标基本恢复正常,Peak分别为(16.60±3.59)mmHg、(16.20±0.23)mmHg、(17.80±4.64)mmHg;dp/dt分别为(742.3±192.1)mmHg、(570.7±77.2)mmHg、(801.0±260.7)mmHg;EDP分别为(0.07±0.54)mmHg (0.69±0.53)mmHg、(1.02±0.18)mmHg;-dp/dt分别为(-523.9±117.3)mmHg、(-464.5±72.8)mmHg、(-632.6±247.6)mmHg,与DM-C组相应指标比较,P<0.05。DM-L、DM-H、DM-rh三组间无显著差别。结论 IGF-I改善STZ-糖尿病大鼠物质代谢同时改善心脏舒张和收缩功能。 【关键词】 糖尿病 心脏功能 胰岛素样生长因子-I
Effects of insulin-like growth factor-I on cardiac function of diabetic rats

【Abstract】 Objective To investigate the effects of recombinant human insulin-like growth factor-I (rhIGF-I) on cardiac function of STZ-induced diabetic rats and compare with these of bovine colostrum truncated IGF-I (BC-tIGF-I). Methods Diabetes was induced in male SD rats (180~220g) by intraperitoneal injection (IP) of STZ at a dose of 55mg/kg and animals were divided into four groups: DM-C group (n=5), treated with NS 0.1ml/d IP; DM-L group (n=5), treated with BC-tIGF-I 250ng.kg-1.d-1, IP; DM-H group (n=5), treated with BC-tIGF-I 750ng.kg-1.d-1, IP;DM-rh group (n=5), treated with rhIGF-I 750ng.kg-1.d-1, IP.
