Balb C乳鼠实验性感染汉坦病毒与脑神经细胞凋亡 |

【摘要】 目的 应用原位末端标记技术对Balb/C乳鼠脑组织的凋亡细胞进行检测,并对脑组织中汉坦病毒(HV)感染和细胞凋亡的关系进行初步探讨。方法 HV陈株经腹腔接种Balb/C乳鼠,感染后分别在1、2、4、8、10和14 d取脑组织,4%多聚甲醛固定,常规制备5 μm石蜡切片,应用原位杂交对乳鼠石蜡切片的病毒定位,用原位末端标记显示乳鼠脑组织中存在着凋亡细胞。结果 原位杂交检测结果显示,病毒主要累及大脑皮层、海马等部位,阳性信号主要分布于胞浆、紧靠核膜,原位末端标记的阳性细胞存在于神经细胞核内,在感染乳鼠脑组织中许多细胞被原位末端标记,呈胞核深染,弥漫分布于皮层、海马、白质等部位。结论 乳鼠脑组织中存在着凋亡细胞,该病变可能与病毒感染有关。 【关键词】肾综合征出血热;凋亡;原位末端标记
Experimental Hantaan virus infection induced apoptosis of nerve cells in the brain of Balb/C suckling mice

【Abstract】 Objective We employed the in situ end labeling (ISEL) to detect the apoptosis in brain nerve cells of the suckling mice to study the relationship between the Hantaan virus in the brain and the apoptosis. Methods Hantaan virus (HV) was injected into the abdominal cavity of the suckling mice. The brain tissue was harvested at 1,2,4,6,8,10 and 14 days after the injection. The specimens were fixed in 4% paraformaldehyde fixative, embedded in paraffin and cut into 5 μm serial sections. We employed the in situ hybridization (ISH) to locate the HV in the brain nerve cells of the suckling mice and detected the apoptosis with ISEL. Results After in situ hybridization, the positive signals were mainly localized in the cytoplasm close to the nuclear membrane in the cells of cerebral cortex, hippocampus. The detection using ISEL revealed the existence of apoptosis in the nuclei of the nerve cells. The darkly stained positive nuclei could be observed scattering in the cerebral cortex, hippocampus, and white matter. Conclusion We found the existence of the apoptosis in nerve cell and probably resulted from the HV infection. 【Key words】Hemorrhagic fever with renal syndrome; Apoptosis; In situ end labeling
