降钙素基因相关肽对内皮细胞增殖及内皮功能的影响 |

【摘要】 目的 观察降钙素基因相关肽(CGRP)对内皮细胞增殖及内皮功能的影响。方法 检测了培养内皮细胞周期时相的分布以及细胞内游离钙离子的水平,同时对培养液中一氧化氮(NO)和内皮素(ET)的含量进行了测定。结果 加用CGRP组,S期细胞明显最多,细胞内游离钙离子水平增高,培养液中NO含量升高,ET含量降低(P均<0.01)。结论 CGRP促进血管内皮细胞增殖,其作用可能与增加细胞内钙离子水平有关,此外,它还能促进NO的合成,降低ET的水平,因此对改善内皮细胞的功能,防治动脉粥样硬化起着积极的作用。 【关键词】 降钙素基因相关肽 内皮细胞 增殖 一氧化氮 内皮素

【Abstract】 Objective To observe the effects of CGRP on cultured endothelial cell.Methods The effect of CGRP on cell cycle kinetics of cultured endothelial cells was investigated by flow cytometry. In addition, the cytoplasmic free calcium turnover was also tested. Nitric oxide (NO) and endothelial (ET), two compounds released by endothelial cell, were tested in order to observe the effect of CGRP on secretion of endothelial cells.Results As compared with control groups, CGRP increased S phase percentage of endothelial cells, the level of cytoplasmic free Ca2+, the NO level, decreased ET level of the conditioned medium.Conclusion CGRP stimulated proliferation of and endothelial cells increased the level of cytoplasmic free Ca2+ indicating an involvement of cyctoplasmic free Ca2+ as second messager in this reaction. In addition, CGRP promoted endothelial cells to secrete NO and also inhibit ET. 【Key words】 Calcitonin gene related peptide Endothelial cell Proliferation Nitric oxide Endothelial
降钙素基因相关肽(calcitonin gene related peptide, CGRP)是迄今为止所发现的体内最强的舒血管活性多肽,其神经纤维广泛地分布于心血管系统[1]。研究表明,CGRP能抑制自发性高血压大鼠以及内皮素介导的鼠VSMC的增殖[2,3],提示CGRP在逆转动脉粥样硬化(AS)及经皮冠状动脉腔内成形术(PTCA)后再狭窄的发生有重要价值,但是有关CGRP对于参与AS和再狭窄发生的另一重要环节——内皮细胞的增殖及功能的影响研究较少。为此我们观察了CGRP对培养的人脐静脉内皮细胞增殖的影响,并检测了细胞内Ca2+的水平以及培养液中一氧化氮和内皮素的含量,旨在进一步揭示CGRP在AS和再狭窄中的价值。
