【摘要】 目的 观察奥曲肽(octreotide, OCT)对人胃癌细胞多种生物学行为的抑制作用,并探讨部分机制。方法 MTT比色分析法测定OCT对胃癌细胞系MKN45生长的影响,流式细胞术检测细胞周期的改变,平板集落实验观察细胞集落形成能力,裸鼠移植瘤形成实验体内观察OCT对胃癌的作用,放免法观察OCT对细胞因子生长抑素(epidermal growth factor, EGF)、转化生长因子-α(transforming growth factor-alpha, TGF-α)生成的影响。结果 OCT能有效抑制MKN45细胞的增殖,呈剂量依赖性,以10-6 mol/L作用最强;OCT诱导肿瘤细胞发生G0/G1期阻滞,以OCT作用24 h时最明显;OCT可显著抑制细胞的集落形成能力,并显著抑制裸鼠移植瘤的形成。OCT作用后细胞分泌EGF、TGF-α的量均明显减少。结论 OCT在体内、体外均能有效抑制胃癌细胞MKN45的生长,可作为胃癌辅助治疗手段之一。

The inhibitory effects of octreotide on proliferation of gastric cancer

Abstract:Objective To investigate the effects of somatostatin analogue octreotide (OCT)on the growth of MKN45 cells and explore its possible therapeutic mechanism.Methods The effect of OCT on cell proliferation in vitro was determined by MTT. Cell cycle was analyzed by flow cytometry. Plate colony formation assay was used to examine the ability of colony formation of MKN45 cells. Tumor transplantation in nude mice was performed to observe the effect of OCT on gastric cancer in vivo. The contents of epidermal growth factor (EGF) and transforming growth factor-alfa (TGF-α) in culture media were measured by RIA.Results The growth of MKN45 cells treated with OCT was obviously inhibited as compared with that of the control. The inhibitory effect of OCT occurred in a dose-dependent manner and was most potent at the concentration of 10-6mol/L. OCT arrested the cell cycle at G0/G1 phrase and reduced colony formation. The repression potential of OCT on the growth of MKN45 cells in nude mice was also noticed. The contents of EGF and TGF-α in the culture media of MKN45 cells were significantly decreased in OCT-treated subgroups as compared with those in the control.Conclusion OCT could significantly inhibit the growth of gastric cancer cell line MKN45 in vi
