【摘要】 目的 观察体外实验中大蒜素对胃上皮细胞系MGC-803 Cx 37 mRNA、Cx 43 mRNA表达及细胞间隙连接通讯功能的影响。方法 终浓度为3 μg/ml、6 μg/ml、9 μg/ml、12μg/ml大蒜素分别加入胃上皮细胞MGC-803培养 24 h、48 h、72 h,同时设立不加药物的阴性对照,用RT-PCR法检测细胞Cx 37 mRNA、Cx 43 mRNA表达,LY染料传输方法检测细胞间隙连接通讯功能。结果 MGC-803细胞Cx 37 mRNA有表达,Cx 43 mRNA无表达,GJIC功能弱,加入不同浓度大蒜素后,Cx 37 mRNA表达和细胞间隙连接通讯功能增强,Cx 37 mRNA表达随大蒜素浓度和培养时间的增加而增强(均P<0.05),Cx 43 mRNA仍无表达。 结论 大蒜素在转录水平上调胃上皮细胞MGC-803 Cx 37基因的表达,改善细胞间隙连接通讯功能。

Effects of allicin on expression of connexin 37,connexin 43 mRNA and gap junction intercellular communication of gastric epithelial cells

[Abstract] Objective To observe the effects of allicin on the expression of connexin 37 mRNA,connexin 43 mRNA and gap junction intercellular communication of gastric epithelial cells MGC-803 in vitro.Methods MGC-803 cell were interfered with various concentration allicin(3,6,9,12 μg/ml ) for 24,48 and 72 hours respectively and control groups with no intervention.RT-PCR method was applied to detect the expression of connexin 37 mRNA,connexin 43 mRNA in cells.Lucifer Yellow Scrape-loading and dye transfer method was applied to detect gap junction intercellular communication.Results MGC-803 cell expressed Cx 37 mRNA,it didn’t express Cx 43 mRNA.Cell gap junction intercellular communication was weak in MGC-803 cell.The expression of Cx 37mRNA and gap junction intercellular communication was enhanced after interfered with variable dose allicin.The expression of Cx 37mRNA was up-regulated with the increasement of concentration and action time of allicin (P<0.05).There was still no Cx 43 mRNA expression in MGC-803 cell.Conclusio
