下肢缺血预适应后大鼠心肌中血红素加氧酶 1与炎症的变化 |

p;male majority Spraguedawley (SD) rats, weighing approximately 200-250 g,were divided into two groups:①AMI group and sham group, ② LIP group and sham group. All the rats both in AMI and LIP groups were sacrificed at 2, 4, 6, 12 h after AMI. Myocardial infarct size was examined with trephine tetrazolium chloride (TTC) staining. Peripheral blood leukocyte count was carried out. Pathological change and the leukocyte infiltrate numbers in myocardium was detected by HEstaining section under high power lens. The serum level of IL8 was determined by using enzymelinked immunosorbent assay (ELISA) and the expression level of IL8 and HO1 mRNA in myocardium were evaluated by method of reverse transcriptionpolymerase chain reaction (RTPCR). Results The myocardial infarct area of LIP group was more significantly decreased than that in AMI group. Inflammatory reaction was significantly less at 12 h after infarct. Conclusions LIP can lessen inflammatory reaction, decrease the myocardial infarct area significantly. It shows that HO1 can protect the infarcted myocardium.
