【摘 要】 目的 了解转化生长因子β1(TGFβ1)在类风湿关节炎(RA)滑膜组织中的表达及与病理改变的关系,以探讨TGFβ1在RA发病中的作用。方法 采用免疫组织化学LSAB法检测了53例(其中26例RA,20例其他关节炎和7例正常人)滑膜组织中TGFβ1蛋白的表达情况,并用目镜网格测微尺和图像分析系统分别进行TGFβ1阳性细胞的半定量计数和TGFβ1染色强度及面积的定量分析。结果 RA滑膜衬里层TGFβ1阳性细胞数及阳性染色强度、衬里下层染色强度及阳性面积均较正常组和其他关节炎组明显增强(P<0.05);RA滑膜衬里层细胞层数、衬里下层炎症细胞的浸润均较对照组明显增多;活动期RA滑膜衬里层和衬里下层TGFβ1染色均较静止期明显增强,并且活动期滑膜炎中,慢性活动期、肉芽组织增生期较新近活动期衬里下层染色强度及阳性面积明显升高(P<0.05)。结论 RA滑膜TGFβ1蛋白合成、分泌增多,并且增多的TGFβ1可能与RA滑膜病变活动有关,包括与衬里层滑膜细胞增生、炎症细胞浸润、衬里下层血管增生及肉芽组织形成有关。 Expression of TGFβ1 in rheumatoid synovia and its relationship with synovial pathological changes

【Abstract】 Objective To know the expression of TGFβ1 in rheumatoid synovia and its relationship with synovial pathological changes for exploring the role of TGFβ1 in pathogenesis of RA.Methods TGFβ1 protein in 53 cases of RA synovia [including 26 cases of RA,20 cases of control non-RA (14 cases of OA and 6 cases of traumatic arthritis) and 7 cases of normal synovia] was detected by immunohistochemical LSAB method.The relationship between expression of TGFβ1 and pathological changes in RA synovia was further analysed.Results ① TGFβ1-positive lining cells and mean greym in intima, mean greym and positive area in sublining area were significantly enhanced in RA synovia than those in control non-RA synovia (P<0.05);② Lining layer and infiltrated inflammatory cells in sublining area were more in RA synovia than those in controls;③ TGFβ1 staining was stronger in active RA synovia especially in chronic active stage and granuloma stage,which showed significantly increased mean greym and positive area in sublining area than that in freshly active stage(P<0.05).Conclusion The results showed enhanced expression of TGFβ1 protein in RA synovia and it may be related wit
