三氧化二砷诱导人胃癌细胞株MGC |

摘 要 【目的】为探讨三氧化二砷(As2O3)对胃癌细胞的作用及可能的机制。【方法】用不同浓度的As2O3作用于培养的胃癌细胞株MGC-803细胞。【结果】噻唑蓝(MTT)细胞活力测定显示As2O3能明显抑制MGC-803细胞生长;进一步用Hoeghes33342和碘化丙啶(PI)双荧光流式细胞仪(FCM)检测和琼脂糖DNA电泳,MGC-803细胞表现为明显凋亡特征;扫描电镜显示凋亡的形态学特征。【结论】As2O3能有效地抑制胃癌细胞的生长增殖,并具有时—效和量—效关系。 关键词 肿瘤细胞,培养的; 胃; 脱噬作用; 砷
Apoptosis of Human Gastric Cancer Cell Induced by Arsenic Trioxide in Vitro

Abstract 【Objective】 To study the effects and possible mechanism of action of As2O3 on human gastric cancer(HGC) cells. 【Methods】 By means of MTT colorimetric assay,the antiproliferative effects of As2O3 with different concentrations on cultured HGC cell line MGC-803 were observed.The characteristics of apoptosis were further examined by Hoeches33342,PI,fluorescence flow cytometry and DNA agarose gel electrophores; whereas the morphological feature of apoptosis was observed by scanning electron microscopy. 【Results】 Prominent apoptosis of HGC cells showing typical morphological changes was induced by addition of As2O3. 【Conclusions】 As2O3 can inhibit significantly the reproduction and proliferation of HGC cells, which is related to drug concentration and time of cultivation. 肿瘤细胞为恶性无限制增殖,分化受阻和细胞凋亡受抑制是恶性肿瘤的重要的发病机制,据此,设计化疗方案,诱导分化和促进肿瘤细胞凋亡是目前肿瘤诊疗的重要措施,特别是促凋亡疗法现已成为各种肿瘤治疗的基础[1]。 近年报道,三氧化二砷(As2O3)能有效地治疗急性早幼粒细胞白血病,并可诱导NB4细胞凋亡[2,3],提示砷剂可能具有抑制某些肿瘤的作用,但有关砷剂对其它肿瘤作用的基础研究尚无报道,砷剂抗肿瘤机制也有待进一步阐明。本研究以人胃癌细胞株MGC-803为受试对象,以观察As2O3能否有效抑制MGC-803细胞生长,促进其凋亡。
