【摘要】 目的 建立反相高效液相色谱法测定小儿清热利肺口服液中总绿原酸的含量。方法 采用DiamonsiLTM C18(250 mm×4.6 mm,5 μm)色谱柱,乙腈体积分数0.4%H3PO4水溶液(体积比10∶90)为流动相,流速为1 mL·min-1,检测波长为327 nm,柱温为30 ℃。结果 绿原酸在质量浓度为16.28~81.4 μg范围内线性关系良好(r=0.999 6,n=5),样品平均回收率为100.31%,RSD为1.45%。结论 本法简便、快速、准确,适用于小儿清热利肺口服液的质量控制。

Determination of total chlorogenic acid in Xiaoer Qingre Lifei oral solution by HPLC

Abstract:Objective To establish a RPHPLC method for the determination of total chlorogenic acids in Xiaoer Qingre Lifei ioral solution. Methods The assay was performed on DiamonsiLTM C18 column with the mobile phase of acetonirile0.4% phosphoric acid solution (10∶90) under a flow rate of 1 mL·min-1. The detection wavelength was set at 327 nm and column temperature was 30 ℃. Results It showed good linearity in the range of 16.28-81.4 μg of chlorogenic acid (r=0.999 6,n=5).The average recovery was 100.31% (RSD=1.45%). Conclusion The method is simple,rapid,accurate and can be used for the determination of total chlorogenic acids in Xiaoer Qingre Lifei oral solution.

小儿清热利肺口服液由银翘散和麻杏石甘汤2个古方合方化裁而成,处方由金银花、连翘、牛蒡子、麻黄、等11味中药组成[1],主治外感风热邪在肺卫或卫气同病之风热咳嗽,用于发热、咳嗽,咯痰、流涕或鼻塞、咽痛、口渴、舌红黄等证候的小儿急性支气管炎。其中绿原酸为金银花的主要活性成分,在牛蒡子中含量亦较高。它是一种具有广泛生理和药理活性的物质,具有利胆、抗菌、降压、增加白细胞及兴奋中枢神经系统、抗肿瘤、降脂、清除自由基等多种药理作用[2]。小儿清热利肺口服液现有质量标准采用紫外分光光度法测定其盐酸麻黄碱含量,并未以绿原酸为含量测定指标。因此,本实验建立高效液相色谱法测定小儿清热利肺口服液中总绿原酸的含量,旨在为完善小儿清热利肺口服液的质量标准提供基础。
