【摘要】 目的 研究骨髓基质细胞的CXCR4表达,SDF-1对BMSCs迁移、趋化作用,分析促进迁移规律和机理。为改善和提高骨髓基质细胞迁移能力寻找更有效的途径。为临床骨髓基质细胞更广泛的应用奠定基础。方法 体外培养大鼠骨髓基质细胞,应用免疫组化方法对骨髓基质细胞的CXCR4受体表达水平进行检测,运用微孔隔离室穿越实验方法,研究SDF-1对骨髓基质细胞迁移、趋化作用,并用SDF-1阻断剂加以证实。结果 CXCR4在各代骨髓基质细胞中表达小于5%,SDF-1可促进骨髓基质细胞迁移。结论 虽然CXCR4在骨髓基质细胞中表达量很少,但是对促进骨髓基质细胞迁移起到至关重要的作用。

The Effects of SDF-1/CXCR4 in Promoting Migration of Bone Mesenchymal Stroma CellsCAO Yang, LV Gang, YU Yang, FAN Zhongkai(Department of Orthopaedics, the First Affiliated Hospital of Liaoning Medical University, Jinzhou 121001 China)Abstract: Objective To investigate CXCR4 expresstion of bone mesenchymal stroma cells, and the effects of SDF-1/CXCR4 in promoting migration of bone mesenchymal stroma cells and to analyze the mechanism of migration of bone mesenchymal stroma cells, which aims to find more effective ways of improving migration ability. Methods Bone mesenchymal stroma cells of rats were cultured in vitro. Receptor of CXCR4 was detected by immunohistochemistry. Migration of marrow stroma cells were investigated by empirical method of transwell. Results The receptor of CXCR4 on bone mesenchymal stroma cells was less than 5%, and SDF-1 could promote migration of bone mesenchymal stroma cells. Conclusions Although there is a little the receptor of CXCR4 on bone mesenchymal stroma cells, which plays an important role in promoting migration of bone mesenchymal stroma cells.

基质细胞衍生因子-1(stromal cel1 derived factor-l,SDF-1)及其趋化因子受体(CXCR4)作为一个趋化因子家族的新成员,在调节造血过程中发挥重要的作用。它在骨髓微环境中的高表达与造血干/祖细胞释放至外周血循环有关,并可作为一个评价造血动员过程中CD34+细胞峰值出现的指标。近来研究表明,SDF-1对多种干细胞和成体细胞有趋化作用,而且是多种干细胞、肿瘤细胞和淋巴细胞的生长因子。因此,随着对SDF-1/CXCR4研究的不断发展,人们愈来愈关注其在多种生物学效应中的重要作用。
SDF-1对细胞的趋化作用是通过其受体CXCR4来调节、是己知的CXCR4的唯一配体。研究表明,SDF-1可诱导大多数循环淋巴细胞和CD34+前体细胞的黏附,使细胞定向性迁移致特殊部位,但SDF-1对CD34-细胞是否有趋化作用尚有争论。本实验目的是运用微孔隔离室穿越实验方法,研究SDF-1对BMSCs迁移、趋化作用,并用SDF-1阻断剂加以证实
