【摘要】 【目的】 用蛋白组学方法,通过对膝关节骨关节炎患者及正常人群血清蛋白的比较,寻找差异表达蛋白并探讨其与骨关节炎致病机理的关系。 【方法】 选取2007年10月到12月10例在中山大学附属第三医院骨科住院治疗的膝关节骨关节炎患者为实验组;10名性别相同,年龄、身体质量指数值相约的健康志愿者为对照组,分析2组双向电泳结果,并对差异表达蛋白进行质谱鉴定。 【结果】 通过双向电泳图普比较发现7个差异表达蛋白,其中6个在关节炎组中呈低表达,1个呈高表达。经质谱鉴定为分别为:6个低表达蛋白分别为对氧磷酶1、对氧磷酶3、α-1-微球蛋白/bikunin前体蛋白、载脂蛋白M、免疫球蛋白轻链、四连接素,高表达蛋白为载脂蛋白L1。【结论】 膝关节骨关节炎血清表达差异蛋白:对氧磷酶1、对氧磷酶3、α-1-微球蛋白/bikunin前体蛋白、载脂蛋白M、免疫球蛋白轻链、四连接素、载脂蛋白L1可能与骨关节炎的发病或病情进展具有一定的相关性。

Abstract: 【Objective】 To seek the differentially expressed proteins in the serum of the patients with knee osteoarthritis compared with normal subjects in control groups by proteomic analysis and to discuss the pathogenesis of osteoarthritis. 【Methods】 Ten patients with knee osteoarthritis, who hospitalized at the Department of Orthopedics, the Third Affiliated Hospital of SUN Yat-sen University from October to December 2007, were selected as the experimental group. Ten normal volunteers with matched gender, similar age, and body mass index (BMI) value were selected as the control group. Two-dimensional electrophoresis (2-DE) patterns of human sera from the 2 groups were analyzed. The differentially expressed spots were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). 【Results】 Seven expression of proteins on dielectrophoretogram were detected to be of significant difference between the two groups. The 6 down-regulated spots were identified as paraoxonase 1 (PON1), paraoxonase 3 (PON3), Alpha-1-microglobulin/bikunin precursor (AMBP), apolipoprotein M (APOM), immunoglobulin light chain (IgL), and tetranectin (TNA).
