婴儿猝死综合征血中抗Ro和抗La自身抗体的检查 |

【摘要】 目的 探讨婴儿猝死综合征(SIDS)的死因。方法 应用重组Ro52、Ro60和La融合蛋白,以ELISA、免疫印迹和免疫沉降法检查了南澳大利亚州的240例SIDS患儿血中的自身抗体。同时取100例成人及32例正常婴儿作为正常对照组。结果 在240例SIDS患儿中,ELISA法发现16.3%具有抗Ro52,7.9%具有抗Ro60,0.8%有抗La自身抗体,部分阳性血清通过免疫印记法加以证明。抗Ro52与Ro60抗体的ELISA阳性率明显高于正常对照组(P分别<0.001和<0.05)。结论 在部分SIDS患儿血中,可能存在来自无症状母亲的抗Ro或(和)La自身抗体,这些抗体可能通过心脏的隐性损伤导致患儿猝死。 【关键词】 婴儿猝死 心脏传导阻滞 自身抗原 核糖核蛋白类
Anti -Ro and anti-La autoantibodies in sudden infant death syndrome ZHAO Yuming*, Gordon T, McCluskey J, et al.* Key Laboratory of Immunodermatology, Ministry of Public Health, the First Hospital of China Medical University, Shenyang 110001 【Abstract】 Objective Sudden infant death syndrome (SIDS) is the most common cause of infant deaths between the age of one week to twelve months in the developed countries. This unexpected death in apparently healthy infants has no known causes. There have been reports on heart conduction system changes as one of the suspected causes of SIDS deaths.Methods Using recombinant Ro60, Ro52 and La fusion proteins with ELISA, immunoblotting and immunoprecipitation as well as routine immunofluorescence and CIE, the authors examined 240 SIDS cases collected in South Australia between 1983 and 1991 for the presence of anti-Ro (SSA) and anti-La (SSB) autoantibodies which are considered to be serological markers of congenital heart block, using 100 normal adult sera and 32 normal infant sera as controls.Results The results showed that about 16.3% SIDS cases had low levels of anti-Ro52, about 7.9% had anti-Ro60 and 2 cases, anti-La (SSB)autoantibodies in their sera as detected by ELISA and verified in some by immunoblotting.
