细胞生长肽对新生大鼠缺氧缺血性脑损伤的保护作用 |

【摘要】 目的 研究碱性成纤维细胞生长肽(bFGF)对新生大鼠缺氧缺血性脑损伤(HIBD)的保护作用。方法 应用105只(bFGF组58只,未治疗组42只,对照组5只)新生Wistar大鼠制备HIBD模型,然后观察bFGF对HIBD模型鼠的体重增长、死亡率、脑损伤的影响,并用原位缺口末端标记(TUNEL)法检测bFGF对缺氧缺血后脑细胞凋亡的影响。结果 bFGF治疗组体重增长为(12±4)g,明显高于未治疗组(6±4)g,差异有非常显著意义(P<0.001);治疗组病死率(5%)低于未治疗组(28%),差异有非常显著意义(P<0.01);治疗组脑病变率(25%)明显低于未治疗组(62%),差异有显著意义(P<0.05)。TUNEL检测结果,治疗组阳性细胞均数(30个/500个细胞)低于未治疗组(159个/500个细胞),差异有非常显著意义(P<0.001)。结论 bFGF对HIBD脑组织有修复和促进细胞生长的作用。 【关键词】 大鼠,Wistar 脑缺氧 脑缺血 脱噬作用 成纤维细胞生长因子,碱性
A study on protective effect of bFGF on hypoxic-ischemic brain damage in neonatal rats SUN Guilian, HAN Yukun. Department of Pediatrics, First University Hospital, China Medical University, ShenYang 110001 【Abstract】 Objective To explore the protective effect of basic fibroblast growth factor (bFGF) on hypoxic-ischemic brain damage (HIBD) in neontal rats.Methods The HIBD models were established with 7 day old Wistar rats. Body weights, mortalities and the degree of brain damage in HIBD rats treated with bFGF were compared with that in HIBD rats without the treatment of bFGF (control group). The effect on apoptosis of neurons was observed by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labelling (TUNEL) staining. Results The body weight in bFGF treatment group was significantly higher than that in control group (12±4) g and (6±4) g, respectively (P<0.001); The death rate in bFGF treatment group (5%) was much lower than that in control group (62%, P<0.01); the average number of positive cells in bFGF treatment group (30/500 cells) was significantly lower than that in control group (159/500 cells) (P<0.001).Conclusion The results verified the protective efficacy of bFGF on HIBD. 【Key words】 Rats, Wistar Cerebral ischemia Cerebral anoxia Apoptosis Fibroblast Growth factor, basic
