孕激素对人卵巢癌细胞株HO8910细胞增殖及凋亡的影响 |

【摘要】 目的 探讨孕激素(P)对人卵巢癌细胞株HO8910细胞体外增殖及凋亡的调控作用。方法 选用人卵巢癌细胞株HO8910进行体外培养,实验组中加入含不同浓度P的培养液,对照组加入等量空白培养液,采用四甲基偶氮唑蓝(MTT)比色法检测细胞增殖,流式细胞仪测定细胞周期和凋亡率,光学显微镜和透射电子显微镜进行形态学观察,终末脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)计数凋亡细胞,流式细胞仪间接免疫荧光技术分析细胞内bcl-2蛋白的表达。结果 P浓度为1×10-7~1×10-5 mol/L时,作用24、48、72 h均明显抑制HO8910细胞的增殖(P均<0.01),并具有剂量依赖性。P作用后G0/G1期细胞增加,并伴随G0/G1期细胞的增加,出现细胞凋亡峰和凋亡率的升高(P<0.01);当P浓度为1×10-6、1×10-5 mol/L时,作用72 h后的细胞凋亡率分别为10.38%和35.78%,两者比较,差异有极显著性(P<0.01)。凋亡细胞计数,当P浓度为1×10-5 mol/L时,8孔中7孔(7/8)为凋亡细胞阳性,明显多于对照组(P<0.05)。光学显微镜和透射电子显微镜可观察到细胞凋亡的形态学改变。细胞内bcl-2 蛋白表达检测显示,当P浓度为1×10-6 mol/L时,bcl-2蛋白表达率为61.25%,明显低于对照组的71.07%(P<0.05);当P浓度增至1×10-5 mol/L时, bcl-2蛋白表达率为9.40%,与对照组和P浓度为1×10-6 mol/L时比较,差异均有极显著性(P<0.01)。结论 P对卵巢癌细胞株HO8910细胞的增殖具有明显抑制作用,呈现剂量-效应关系。P的这种抗癌作用与诱导细胞凋亡有关,抗凋亡基因bcl-2 蛋白表达下降可能是诱发细胞凋亡的机理之一。 【关键词】 卵巢肿瘤; 孕酮; 囊腺癌,浆液; 脱噬作用; 肿瘤细胞,培养的
The Effect of Progesterone on Proliferation and Apoptosis in Ovarian Cancer Cell

【Abstract】 Objective To investigate the regulatory effect of progesterone on proliferation and apoptosis in ovarian cancer cell line HO8910 in vitro. Methods Ovarian cancer cell line HO8910 originated from human ovarian serous cystadenocarcinoma was cultured in vitro. Two groups were set up: study group (progesterone in different concentrations) and control group without progesterone. Cell proliferation was measured by 3-(4,5-dimethylthiazol-z-yl)-2,5-dipheny tertrazolium blue (MTT) colorimetric assay. Cell cycle and apoptotic percentage were detected by flow cytometry, morphological changes of apoptotic cells were observed by light and electron microscopy, and apoptotic cells were quantitatively determined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL).
