雌激素对卵巢癌细胞株体外生长的影响 |

【摘要】 目的 探讨17-β雌二醇(E2)对卵巢癌细胞株体外生长和细胞周期的影响。方法 采用四甲基偶氮唑蓝(MTT)比色法和流式细胞术(FCM)的方法,观察了卵巢癌细胞株CAOV3和OVCAR3在加入含有不同浓度17-β E2培养液后细胞活性和细胞周期的变化;同时采用免疫组化方法,测定癌细胞在加入17-β E2前后雌激素受体表达的变化。结果 (1)浓度为0.25~5.00 nmol/L的17-β E2,可轻度抑制卵巢癌细胞株的生长,抑制率为6.3%~36.0%。(2)17-β E2可使CAOV3细胞株中G0及G1期的细胞由55.0%减少为19.0%~30.0%,S期的细胞由30.0%增加到52.0%,G2及M期的细胞由14.0%增加到17.0%~28.0%;使OVCAR3细胞中G2及M期的细胞由37.0%减少为22.0%~30.0%,S期的细胞由31.0%增加到38.0%~51.0%。17-β E2对G0及G1期的OVCAR3细胞没有明显的影响,加入17-β E2前为32.0%,加入17-β E2后为29.0%~31.0%。(3)卵巢癌细胞株CAOV3和OVCAR3均为雌激素受体阳性细胞株,加入17-β E2对其雌激素受体的表达没有明显影响。 结论 雌激素对卵巢癌细胞株CAOV3 和OVCAR3没有刺激其生长的作用, 相反可轻度抑制CAOV3和OVCAR3细胞的生长,可能是雌激素通过细胞的雌激素受体改变了细胞的生长周期。 【关键词】 卵巢肿瘤;雌激素替代疗法;雌二醇
Influence of Estrogen on the Epithelial Ovarian Cancer Cell Lines in Vitro

【Abstract】 Objective To study the effect of 17-β estradiol on the growth of the epithelial ovarian carcinoma cell lines in vitro. Methods The proliferative capacity of the ovarian carcinoma cell lines CAOV3 and OVCAR3 in the cultrue medium with 17-β estradiol was evaluated by the means of the mircocultrue tetrazolium assay (MTT) and cell kinetics was analyzed by flow cytometry (FCM). The expression of estrogen receptor (ER) of the two cell lines was examined by the immunohistochemical staining. Results The growth of both two epithelial ovarian cancer cell lines was slightly inhibited by 17-β estradiol (0.25~5.00 nmol/L), and the inhibitive rate was 6.3%~36.0%. FCM showed that the kinetics of the two cell lines were obviously changed.
