【摘要】 目的:探讨COX-2抑制剂NS-398对转化生长因子-β(TGF-β)影响肾小球系膜细胞表达环氧化酶-2(COX-2)及Smad2 mRNA表达的作用。方法:传代培养系膜细胞后取第4代系膜细胞进行分组实验,用RT-PCR方法测定各组COX-2 mRNA及Smad2 mRNA的表达,观察不同浓度TGF-β在4、12、24 h对COX-2 mRNA及Smad2 mRNA表达的影响,及NS-398对其的抑制作用。结果:不同浓度TGF-β作用不同时间后,系膜细胞COX-2 mRNA和Smad2 mRNA表达均增加,呈浓度和时间依赖性。与TGF-β组相比,加入COX-2抑制剂NS-398后,COX-2 mRNA和Smad2 mRNA表达均减少。结论:TGF-β刺激肾小球系膜细胞COX-2表达的信号通路之一是Smad信号通路。而COX-2抑制剂NS-398对COX-2表达的抑制机制之一是通过此信号通路。

[Abstract] Objective: To investigate the effects of inhibitor of COX-2 on the expression of cyclooxygenase-2 by transforming growth factor β and Smad signaling pathway in cultured rat mesangial cells. Methods: Using the quart glomerular mesangial cell for experimentation, to observe the effect of different concentrations of TGF-β on the expression of COX-2 and Smad2 mRNA at 4, 12 and 24 hours, and the effect of the inhibitor of COX-2 NS-398 on the expression of COX-2 and Smad2 mRNA. The expression of COX-2 mRNA and Smad2 mRNA was determined by RT-PCR. Results: After the glomerular mesangial cell was irritated by TGF-β, the expression of COX-2 mRNA and Smad2 mRNA increased. This depended on the density of TGF-β and the length of TGF-β acting time.
