抑制一氧化氮合成对孕鼠血管调节因子的影响 |

【摘要】 目的 研究抑制一氧化氮(NO)合成对孕鼠血浆内皮素(ET)、血栓烷素(TXA2)、前列环素(PGI2)及血管紧张素II(ANG II)水平变化的影响;并探讨这些因子在妊高征发病中的作用。方法 选择清洁级Wistar大鼠,随机分为两组,(1)对照组12只,于妊娠第14天开始皮下注射生理盐水直至分娩。(2)亚硝基左旋精氨酸甲酯(L-NAME)组,12只,于妊娠第14天开始皮下注射一氧化氮合酶(NOS)抑制剂L-NAME,每日125 mg/kg体重,直至分娩。测定两组孕鼠尾动脉收缩压、尿蛋白、血小板数及胎鼠胎盘重量。测定两组血浆NO、ET、PGI2、TXA2及ANG II水平。结果 L-NAME组孕鼠出现类妊高征特征,表现为血压明显升高至(140.6±3.8)mm Hg(1mm Hg=0.133kPa),尿蛋白含量增加为(801.38±57.12)mg/L,血小板数显著减少至(533±42)×109/L,及胎鼠胎盘重量明显减轻。对照组孕鼠血压(108.6±3.4)mm Hg,尿蛋白(413.22±85.55)mg/L,血小板计数(876±34)×109/L。两组比较,差异有显著性(P<0.05)。与对照组比较,L-NAME组孕鼠血浆NO、PGI2和ANG II水平明显降低,而血浆ET及TXA2明显升高。两组比较,差异有显著性(P<0.05)。结论 抑制NO合成可使孕鼠体内血管舒张因子减少、血管收缩因子增多;妊高征的发生可能是体内血管调节因子平衡失调所导致的结果。 【关键词】 妊娠并发症,心血管 高血压综合征 一氧化氮 内皮缩血管肽类 依前列醇
The Effect of Inhibition of Nitric Oxide Synthesis on Vasoregulatory Factors in Pregnant Rats

【Abstract】 Objective To study the effect of the inhibition of nitric oxide(NO)synthesis on plasma endothelin (ET),prostaglandlin I2(PGI2),thromboxane (TXA2)and angiotensin (ANGII) in pregnant rats, and to investigate the role of these factors in the pathogenesis of pregnancy-induced hypertension(PIH). MethodsPregnant rats were divided into two groups randomly.Saline solution or L-nitro arginine methyl ester (L-NAME) 125 mg/day was given subcutaneously from day 14 of gestation till delivery.Systolic blood pressure, urine protein, platelet count,and weight of pups and placentae were determined. Plasma NO, ET, PGI2, TXA2 and ANGII were measured by RIA.
